Im trying to Italicize a single word of text in a lower third that has several sentences contained within the graphic. How does this work in the sequence mode?

You'll need to create a different font that is italicized. If the text object is not published then you can highlight the word in layout and double click the font with italics. When played back from the sequencer this font choice will be maintained.

If your text object is published so that it can be changed from the sequencer; then you need to apply the font in the Template Data tab in the sequencer item. Just put your cursor before the word you want in italics and click the icon with the "aA" above the text.. This will open a dialog where you can choose a font.. You'll see it applies a font tag into the text to indicate the desired font.

For some reason in the layout mode Under the Arial font, Italics is not an option, the box is grayed out? If I change the font to Calibri then the box can be changed to bold Italics? Why cant I change the Arial font to bold Italics?

Sounds like you don't have that option for Arial. I don't have it on my machine either. Some fonts come with multiple options and others do not.

But, you do have a SLANT option in your Scene Fonts Tab that creates a "italic" look to any font.
