First you have to add the Layouts as Sources in your database.
Then make sure you have your Heads as destinations. They may have be added at database creation but you might have to add those too.
If you are using soft panels to route, you will need to add both the new layout Source to your panels.
In Salvos you now can send a layout to a head.
There are some instructional videos on YouTube that show it step by step including a very important Routing Devices Refresh button that had me stuck for a while.

Ultrix 6.4.
I have salvos that assign a layout to an UltriScape head in addition to assigning the sources to the PiPs.
Some of the PiPs need to be Destination Follows.
Is there a way to include Destination Follows in my MV / PiP salvos?
The fastest way I have found is to fire the Salvo then reassign the layout via UltriScape Head Selection with the Destination Follows applied.
I thought about building unique layouts with higher unused PiP numbers permanently defined as Destination Follows under Configurations.
Is there a better way?
