sorry I just figured that in the video coder (I used 9.7) with the mp4 format of your files you could not use "The Alpha from Video fill" and the Alpha from RGB did not work eather with what you needed.
So what you need is first just the video mask - and for that a Xpression AVI with the Alpha Channel having the black and white information.
Do you have After Effects? Do you know how to do that?
None of your pictures need to be the fill (source) in this case the fill does not matter because you use just the alpha as the mask information.
Then to archive what you want there are 2 ways:
1. build a scene that has the transition - means the mask video in a layer and the picture quad published with material. Then in the sequncer you have to put them always a layer higher - so bothe scenes will be onair the one with the old picture and the one with the new picture - once the new scene is onair then take the scene before offline. Not very practicle handlingwise but o.k. for just a few pics.
or better:
1 scene with 2 pictures - the old and the new both published. The old picture is always the one from the scene before.
Demoprojekt attached and later I will make a little explanation how to archive a different alpha in After Effects.

From the tutorial you provided, if I want to create a transition effect like the one I'm trying to achieve, should I register the video source in the brush transition in the Xpression Video Codec and register the desired video image in the alpha source?

What I want is for the second video to appear with a brush transition effect applied when transitioning from the first video.

incase you just want to use your black and white video as a reveal or transition you need:
1. have the black and white information in the alpha channel of your video - video tutorial B124

or possible in After Effects with having the material as a luminanz alpha
2. just ad material -> then mask -> then video
It actually does nothing else then setting all the blending modes automaticly like in the pic. above.
3. Create a layer in the scene and put your video mask and the material etc. you want to reveal in the layer - be aware of Z-achsis or position above or under with the mask to work.
The mask video can be draged in the scene director or have a start automaticly - all works.
From what I got above I think you had the black and white informatin only in the fill and not in the key (alpha) of the video.
Attached a example - incase I understood what you want to do wrong just mail me directly what you mean.

Only because I know it is a really busy time for all Ross staff with NAB happening in America, I wanted to get you a response. What you are asking for is possible. You will have to change blending modes in the material manager to make this video file an alpha. - Then if you want to wipe inside of a scene, you probably not only need to bring the video into your project as a material, but you also need to bring that video file into a scene director.
